1990年代，上海某所高校。来自偏远小城的杨铮（李亚鹏）自小好强，大学里亦不例外。他结识不同专业但同爱流汗的乐言（王学兵）、雨森（廖凡）、佳伟（崔达治）和若彤（王渝文）后，四个男生组成了一支男子接力赛跑队伍，若彤做了拉拉队的队长。随后如天使的女孩文慧（徐静蕾）走进杨铮的生活，成为他生命中最重要的人，他不知文慧是雨森暗恋长达一年的女孩，也不知若彤一直对他倾心，而乐言又偷偷爱着若彤，但这些没有影响他们的友情，几个年轻人在操场上度过了许许多多的欢乐时光。
激情褪去，杨铮与文慧的爱情遭遇生活。杨铮要毕业时，文慧与他就工作问题发生激烈的争吵，两人分手，杨铮去了南方做生意，伤心的文慧开始与雨森交往。雨森察觉到杨铮依旧在文慧心中占有很重的分量时，动情向她表白，却不幸死于车祸，杨铮返校与大伙一起以跑接力的方式将他怀念，并借机对文慧重新展开追求，文慧无法接受。不久文慧之外的所有人毕业离校，若彤进了电视台做记者，乐言成了设计师，杨铮则又回南方。
一年过去，始终放不下文慧的杨铮再返校园，却得知文慧即将赴美，伤心欲绝。不久他在投机生意中被采访的若彤发现，震惊之余，若彤极力相帮，没能改变他被判入狱一年的命运。期间乐言结识精灵女孩小艾（谢雨欣），两人与若彤等人在心烦意乱时仍会像大学时那样在操场上借助奔跑、呐喊将之赶走。杨铮出狱后几乎一无所有，若彤成了帮助他重建自信的身边人，杨铮对她慢慢由感激转为爱意，这时文慧归国。
